Methyl Green Coutnerstaining Procedure
Reagents:
0.8% Methyl Green (Sigma M5015) Sodium Acetate (Sigma
S9513)
Recipe:
Dissolve 1.36 grams Sodium Acetate in 80 ml Di H2O.
Adjust the pH to 4.0 with Acetic Acid. Add Di H2O to a final volume
of 100 ml. Dissolve 0.8 grams Methyl Green in the 100 ml. Filter through
a 0.45 or 0.2 micron filter prior to use.
Procedure:

**It should stain normal nuclei a pale to medium green if the pH is correct
and the methyl green does not contain any crystal violet
**Not suitable for AEC stained slides
