Schneider Lab Gelatin Embedding (Bjorn Merker)

 

Solution A.

90 g. Albumen
100 cc. H2O
Stir with a stick to mix thoroughly

Solution B.

9 g. Gelatin
200 cc. H2Q (warm)
Mix on electric stirer

Pour B into A and stir. Knead and then strain through a net to get smooth.
Store in refrigerator. If mold forms after a few weeks it is not necessary to discard. Scrape mold off top.

Medium will solidfy in the frige. To embed liquify it in a warm water bath.

1. Prepare cardboard casette with a piece of glass on the bottom.
2. Take well fixed brain from sucrose solution.
3. Rinse brain for approximately 20 seconds to remove sucrose from the surface.
4. Blot brain thoroughly with a Kimwipe.
5. Cover brain with medium intimately using the corner of a Kimwipe.
6. Place a little medium over the bottom of the boat with a dropper.
7. Put brain into boat and stabilize in desired orientation with pins
8. Fill boat down one corner with medium using the dropper.
9. Set in air approximately 15 minutes until a skin froms.
10. Expose boat to formaldehyde vapors. Prevent it from comming in contact with the liquid.
11. Set 12-24 hours.
12. When you touch the surface and nothing comes off on your finger a real skin has formed. At this time place boat into stock formalin for three days.
13. After peeling boat away and removing glass and pins section on freezing microtome.
