CELL DISSOCIATION -SCHMIDT. BJORKLUND & STENEVI

1. Collect tissue from 9-12 fetuses over 30-60 min period in room temperature sterile saline or sterile saline supple- mented with 0.6% D-glucose.
2. Incubate tissue at 37 degrees C. for 20 min in 150-200 ul sterile saline containing 0.6% D-glucose and 0.1% trypsin.
3. Wash off trypsin by replacing fluid with room temperature saline-glucose solution 4 times.
4. Disrupt tissue by repeated pipetting through a fire-polished Pasteur pipette.
5. 5. Store solution up to 2 hours at room temperature in saline- glucose solution.
